The Mighty Quick Protocol offers our customers the advantage of quality antibody titers in less time than is required in our traditional protocols. The Mighty Quick Protocol utilizes a proprietary reagent we call Mighty Quick Stimulant that accelerates the humeral immune response. The Mighty Quick Stimulant does not replace conventional adjuvants; it performs in conjunction with conventional adjuvants. With our specialized immunization techniques and optimized injection site placement, your antigen receives the best possible treatment for superior quality antibodies

The Mighty Quick Stimulant shows increase in antibody related cytokines at the protein and mRNA levels. Subsequently, there is an enhanced maturation of antibody producing B-cells and their transformation into antibody producing plasma cell. The Mighty Quick Stimulant induces an earlier switch in IgG isotopes from IgM to IgG1 and IgG2. Finally, the Mighty Quick Protocol has successfully induced this increased response all seven of the species offered by Pocono Rabbit Farm & Laboratory.

These antibody responses at day 28 have been shown as equal or superior to the response that we see at day 70 in our 70 Day Antibody Production Protocols. We inject an Immune Stimulator along with the standard antigen preparation, along with an accelerated immunization schedule. We combine our proven technique with a new immune stimulator to get the same antibody faster.

HOW DOES IS WORK?

When the Immune Stimulator is injected into animals it stimulates induction of cytokine cascades which closely resemble cytokine cascades, as seen during early stages of immune response. As a result, the animal is more prone to producing a faster and strong immune response.

WHAT IS THE IMMUNE STIMULATOR?

The Immune Stimulator is a proprietary product. However, we can tell you that it is a 2.5 kD molecule that, when injected into animals, induces cytokine cascades that lead to activation of immune cells involved into humoral immune response to alien antigens. This makes the animal more responsive to the antigen. Traditional adjuvants, such as Freunds, work by changing the immunogenicity of the molecule or the way it is presented to immune system.

ARE TRADITIONAL ADJUVANTS USED?

YES, both the Immune Stimulator and traditional adjuvants are used together because both have different functions. The Immune Stimulator has been used with Freunds Complete, Freunds Incomplete, Titermax, and Pam3cys.

IS IT SAFE FOR THE ANIMALS?

The Immune Stimulator is safe for the animals. In our experience, we have found no adverse reactions.

HOW MANY INJECTIONS WITH THE IMMUNE STIMULATOR?

For the 28 day protocol; three injections. For the 49 day protocol; four injections. When the project is extended for longer periods, we boost with traditional methods.

I HAVE TRIED UNSUCCESSFULLY TO MAKE ANTIBODIES TO A ….

The Immune Stimulator will not make non-antigenic antigens produce antibodies. If your antigen has not made antibodies in a traditional protocol, it will not make antibodies with the Mighty Quick Protocol. If you feel your antigen is a poor candidate for antibody production and you still want to try, we suggest using our 70 Day Antibody Production Protocols.

No, the Immune Stimulator is not for sale.

28 DAY MIGHTY QUICK PROTOCOL - IN RABBITS

DAY 0 - Preimmune Bleed, 3 to 4ml of sera. Preimmune bleed is stored until the day 21 and day 28 bleeds are sent.

Inject 200ug intradermal with Mighty Quick Immune Stimulator in Complete Freund's Adjuvant (FCA). Antigen supply should be 1mg/ml or higher. If the antigen is not suitable for intrdermal injections, the rabbit will be injected subcutaneous.

DAY 7 - Inject 100ug subcutaneous with Mighty Quick Immune Stimulator in Incomplete Freund's Adjuvant (IFA)

DAY 14 - Inject 100ug subcutaneous with Mighty Quick Immune Stimulator in IFA.

DAY 21 - Large Bleed, about 15ml of sera. This bleed is stored until day 28.

DAY 28 - For CLOSED projects - The rabbit is exsanguinated (final bleed) and the sera sent you, along with the day 21 bleed and the preimmune bleed

For OPEN projects - Large Bleed - This bleed is then sent to you, along with the day 21 bleed and the preimmune bleed.

AFTER DAY 28 FOR OPEN PROJECTS - You may select one of the project extensions below. If you do not specify a project extension, we will automatically begin Extension A. Keep in mind that all procedures and maintenance after day 28 will be charged according to the Custom Polyclonal Antibody Production Price Schedule. Only one immunization after day 28 will include the Mighty Quick Immune Stimulator. This is generally the day 35 immunization.

EXTENSION A: We will boost every four weeks with 25-50ug of antigen subcutaneous with IFA, starting at day 35. We will do a large bleed two weeks after each boost and ship. We will continue to boost and bleed every four weeks until you contact us.

EXTENSION B: When the antisera contains sufficient antibody titer and large volumes of sera are needed, we can start production bleeds. We do production bleeds every seven days. We will continue with production bleeds until you contact us or the health of the rabbit requires us to stop production bleeds. The volume of serum collected for large or production bleeds depends on the weight and the general health of the Rabbit. We generally take 25-45ml of blood yielding 13 to 20ml of serum. During this time, we boost the rabbits every four weeks with 25-50ug of antigen subcutaneous with IFA. You may also request that we not do any boosts after the initial immunization scheme outlined above since some antigens and some animals may respond negatively to boosts during the period of peak antibody activity. Boosts may be required only when titer drops. It is up to the investigator to decide and let us know if we should discontinue boosts as our standard schedule provides for boosts every 28 days. Shipping for production bleeds is every four weeks unless you request otherwise.

EXTENSION C: Some investigators prefer to "wait and see." after day 28. Extension C is hold without boosting and bleeding. We will hold until you tell us to do something else.

TERMINATION: When you have collected enough antisera, you will need to contact us to terminate the project. You may request that we terminate by:
1. Exsanguination - termination with sera - the rabbit is anesthetized and the terminal bleed is taken. For best results, rabbits should be boosted 10 to 21 days before exsanguination.
2. Discard - termination without sera - the rabbit is humanely euthanized.

When requesting termination, you must confirm the species and animal number. Written confirmation by email or fax may be requested for terminal procedures.

28 DAY MIGHTY QUICK PROTOCOL - IN RABBITS
